Annexin V is a Ca2+-dependent phospholipid-binding protein with a molecular weight of 35kD to 36kD. It binds specifically to phosphatidylserine (PS) flipped outside the membrane during apoptosis. In normal cells, phosphatidylserine (PS) is only distributed on the inner side of the lipid bilayer of the cell membrane. But in the early stage of apoptosis, the phosphatidylserine (PS) in the cell membrane turns from the inner side to the outer side. Annexin V labeled with fluorescein is used as a fluorescent probe to detect the occurrence of apoptosis by flow cytometer or fluorescence microscopy.
